Vallone, Daniela, Elena Frigato, Cristiano Vernesi, Augusto Foà, Nicholas S. Foulkes, and Cristiano Bertolucci. Hypothermia modulates circadian clock gene expression in lizard peripheral tissues.Am J Physiol Regul Integr Comp Physiol 292: R160–R166, 2007. First published June 29, 2006; doi:10.1152/ajpregu.00370.2006.— The molecular mechanisms whereby the circadian clock responds to temperature changes are poorly understood. The ruin lizard Podarcis sicula has historically proven to be a valuable vertebrate model for exploring the influence of temperature on circadian physiology. It is an ectotherm that naturally experiences an impressive range of temperatures during the course of the year. However, no tools have been available to dissect the molecular basis of the clock in this organism. Here, we report the cloning of three lizard clock gene homologs (Period2, Cryptochrome1, and Clock) that have a close phylogenetic relationship with avian clock genes. These genes are expressed in many tissues and show a rhythmic expression profile at 29°C in light-dark and constant darkness lighting conditions, with phases comparable to their mammalian and avian counterparts. Interestingly, we show that at low temperatures (6°C), cycling clock gene expression is attenuated in peripheral clocks with a characteristic increase in basal expression levels. We speculate that this represents a conserved vertebrate clock gene response to low temperatures. Furthermore, these results bring new insight into the issue of whether circadian clock function is compatible with hypothermia.

Nitric oxide in the nucleus raphe magnus modulates cutaneous blood flow in rats during hypothermia

Objective(s): Nucleus Raphe Magnus (NRM) that is involved in the regulation of body temperature contains nitric oxide (NO) synthase. Considering the effect of NO on skin blood flow control, in this study, we assessed its thermoregulatory role within the raphe magnus. Materials and Methods: To this end, tail blood flow of male Wistar rats was measured by laser doppler following the induction of ...

C H IN A Synchronization of peripheral circadian clocks in rats by external time cues

Circadian clocks are present in a variety of tissues and cells in mammals and appear to be organized in a hierarchical manner. They are divided into a master clock that is localized in the hypothalamic suprachiasmatic nucleus (SCN) and peripheral clocks in extra-SCN brain regions and peripheral tissues, such as heart and liver. The circadian clock is generally reset by environmental time cues, ...

Sympathetic input modulates, but does not determine, phase of peripheral circadian oscillators.

The circadian clock in the suprachiasmatic nucleus (SCN) maintains phase synchrony among circadian oscillators throughout the organism. Environmental light signals entrain the SCN, but timed, limited meal access acts as an overriding time cue for several peripheral tissues. We present data from a peripheral oscillator, the submaxillary salivary gland, in which temporal restriction of meals fail...

Ketogenic diet and fasting induce the expression of cold-inducible RNA-binding protein with time-dependent hypothermia in the mouse liver☆

Cold-inducible RNA-binding protein (CIRBP) induced by cold stress modulates the molecular circadian clock in vitro. The present study examines the effect of a ketogenic diet (KD) and fasting on Cirbp expression in the mouse liver. Chronic KD administration induced time-dependent Cirbp expression with hypothermia in mice. The circadian expression of clock genes such as Bmal1 and Clock was phase-...
